Abstract: The talk will cover both siRNA and shRNA-based technology, focusing on how to increase the knockdown efficiency and reduce the off-target effect that may lead to toxicity or false phenotypes at the same time. Also we will present results on effective gene silencing even in difficult to transfect cells such as neuronal, suspension and stem cells without a transfection reagent, electroporation techniques or viral delivery. Recently, numerous publications showing that RNAi screening strategies have been applied in the studies of signal transduction, cancer progression and host cell responses to infection. Large siRNA, shRNA and microRNA genome-scale libraries screening applications in these biological processes studies will also be introduced. Thermo Fisher Scientific is a company with leading RNAi technology which comprises different well-known product lines like Dharmacon and Open Biosystems.
